小麦幼叶中小G蛋白的粗分离

The Crude Isolation of Small G Protein from Young Leaves of Wheat

摘要 对小麦幼苗叶片中的蛋白进行了粗提,聚丙烯酰胺凝胶电泳(SDS-PAGE)检测在20.1 KDa~31 KDa间呈现一条蛋白条带,经分析其分子量约为23.1 KDa,占所提取总蛋白的2.93%,推测其为小G蛋白家族中的成员.采用传统的硫酸铵盐析法对此目的蛋白进行粗分离.结果显示,30%的硫酸铵饱和度可将该蛋白分离,且所含杂蛋白量最少,其纯度和得率分别是22.96%和47.3%,是进一步细分离纯化的较好选择. The proteins in wheat seedling leaves was crudely extracted.SDS-PAGE clearly revealed a protein band between 20.1 KDa and 31 KDa,its molecular weights was about 23.1 KDa,contented 2.93% of total protein.We sepculated that it belongs to the family of small G protein.The salt precipitation assay was used to primary separate this protein as objective protein.The results showed that this protein can be separated by 30% saturation of ammonium sulfate,containing the least other proteins,the yieldrate is 47.3% and purity is 22.96%,that is the better choice for further purification.
