摘要
目的 了解烟草浸润液对人成骨细胞在钛板上的黏附数量、形态、增殖能力及成骨能力的影响.方法 以不同浓度的烟草浸润液作用于钛板上黏附的人成骨细胞,倒置显微镜观测细胞形态的变化,MTT法测定成骨细胞的钛板黏附数量和增殖能力,磷酸苯二钠法测定成骨细胞碱性磷酸酶分泌情况.结果 随着ST浓度的增加,细胞伸展的范围逐渐缩小,形态逐渐呈现为长条形,细胞在钛板的黏附数量、增殖能力及碱性磷酸酶分泌量成浓度依赖性抑制.结论 ST不利于钛-骨间形成良好的骨整合,是导致种植失败的重要因素之一.
Objective To investigate the effect of smokingless tobacco extract (ST) on munber, adhesion, morphology and proliferation of human osteoblasts on titanium implants in vitro. Methods Human osteoblasts on titanium implants were cultured in presence of ST at various concentrations. The morphologic changes of osteoblasts with different concentration of ST were observed under the inverted microseope. The proliferation and attachment of cells were analyzed by MTT method. The activity of alkaline phosphatase(ALP) was measured by di-sodimn phenyl phosphate method. Results The size of cells became smaller and their shape changed when the concentrations of ST increased. The inhibition of growth, adhesion and ALP activity was concentration dependent. Conclusion The ST is a disadvantage factor for the osseointegration between the bone and titanium implant interface and is also main factor of failure in implantation.
