摘要
目的:采用薄层色谱(TLC)法及高效液相色谱(HPLC)法分析贵州产菝葜属(SmilaxL.)、肖菝葜属(HeterosmilaxK.)8个品种中的活性成分——白藜芦醇,并通过对8个品种间镇痛试验的对比研究,对品种间进行药效与质量的评价。方法:TLC法用硅胶G板,以氯仿-丙酮-醋酸-水(4:4:0.5:0.2)为展开剂定性分析。用HPLC法分析白藜芦醇,色谱条件:依利特HypersilODS2色谱柱(4.6mm×250mm,5μm),流动相为乙腈-0.05%磷酸(24:76);流速0.8mL·min-1;检测波长306nm,理论塔板数以白藜芦醇计不得低于4000。用冰醋酸所致小鼠扭体影响进行镇痛试验。结果:在TLC鉴别中,8个品种均含有白藜芦醇;在选定的色谱条件下,白藜芦醇在0.0062~0.2004μg范围内与峰面积呈良好线性关系。加样回收率(n=9)为101.07%,RSD为1.13%。8个品种中白藜芦醇含量为0.0060%~0.0439%,其中以小叶菝葜含量最高。通过镇痛试验,显示8个品种均有明显镇痛作用,且在相同有效剂量下,8个品种间没有明显差异。结论:本法的TLC法重复性好,专属性强;含量测定方法简单易行,准确度高,可为菝葜属及肖菝葜属新的药用资源开发利用及质量控制提供参考依据。
OBJECTVE Established TLC and RP-HPLC methods to analyze the the active constituents resveratrol contained in 8 variety from Smilax L. and Heterosmilax K. in Guizhou province. Through out Analgesic experiments to evaluate the analgesic effect and quality between 8 varieties. METHODS The TLC qualitative analysis carried out with silica gel G plate and with the CHCl3-CH3 COCH3-CH3 C(X)H-H20 (4 : 4 ;0. 5:0. 2) as the developing solvent. Resveratrol was determined by RPHPLC method, column: elite Hypersil ODS2 (5μm, 4. 6×250mm), mobile phase: CH3CN-0. 05 % H2PO4 (24: 76) ; flow rate: 0. 8 mL-min-1 ;detection wavelength:306 nm,The theoretical plate number of resveratrol cannot be less than 4,000. Acetic acid-in- duced writhing test analgesic effects. RESULTS In the TLC identification,8 varieties contain resveratrol; in the selected chro- matographic condition, the resveratrol showed good linear relationships with the peak area in the range of 6. 3 - 200. 4/zg. Tbe recovery rate of resveratrol (n = 9) was 10.07%, RSD = 1.13%. 8 species' resveratrol content range was (1. 006 0- 0. 043 9%, of which the highest content was S. microphylla. Through analgesic study, all of the 8 varieties had significant anal- gesic effects,and there was no significant difference among varieties at the same effective dose. CONCLUSION The TLC method in this study is high accuracy and reproducible and the RP-HPLC method is simple, specific, they can provide some ref- erence for new medicinal resources development and quality control of Smilax and Heterosrnilax plants.
