摘要
目的比较BALB/c小鼠在幽门螺旋杆菌(Hp)感染后和用益生菌治疗后胃黏膜内Toll样受体4(TLR4)、MyD88和IL-1β在基因和蛋白水平上的表达变化,探讨Toll样受体4信号通路在益生菌治疗Hp感染中的作用。方法 60只感染Hp的BALB/c小鼠随机分为3组,第1组不予治疗,第2组予抗生素三联疗法治疗,第3组予乳酸杆菌治疗,另设20只正常小鼠为对照组。取各组小鼠的胃黏膜标本,用RT-PCR和Western blotting的方法半定量检测TLR4、MyD88、IL-1β在基因和蛋白水平上的表达。结果①正常小鼠胃黏膜内无TLR4、MyD88和IL-1β表达。②益生菌治疗组小鼠的TLR4、MyD88和IL-1β均高于抗生素治疗组,但二者均低于Hp感染组。③各组小鼠TLR4与MyD88的表达均呈正相关。结论 TLR4信号通路参与了Hp的致病机制和益生菌治疗Hp的机制,其信号转导均可能是通过MyD88途径。
[ Objective ] To compare expression of TLR4, MyD88, IL-1β on gene and protein level between after Hp infection and after treatment with probiotics in BALB/c mice. To investigate the role of TLR4 signal pathway in probiotie treatment of Hp infection. [Methods] 60 Hp infected BALB/c mice were randomly divided into 3 groups, Group 1 untreated, Group 2 treated by triple therapy of antibiotic, group 3 treated by Laetobaeillus, a separate 20 mice were the control group. RT-PCR and Western blotting were used to semi-quantitative detect of TLR4, MyD88, IL-1β on gene and protein level in the gastric mueosa of mice. [Results] 1. There was no expression of TLR4, MyD88, IL-1β in the gastric mucosa of normal mouse. 2. Expression of TLR4, MyD88, IL-1β in probiotic treated mice group were higher than the antibiotic treatment group, but both were lower than Hp infection group. [ Conclusions ] TLR4 signal pathway is probably involved in the mechanism of probiotic treatment of Hp infection.
