摘要
目的:研究不同促渗剂对马钱子碱贴剂体外透皮促渗作用的影响。方法:采用不同浓度、不同种类促渗剂制备马钱子碱贴剂;采用改良Franz扩散池,以离体雄性大鼠皮肤为模型,通过高效液相色谱法测定药物浓度,拟合马钱子碱透皮吸收的累积透过量和透过速率。结果:以3%氮酮制备的马钱子碱贴剂具有较好的体外透过速率及累积透过量;促渗剂合用时以3%氮酮+3%薄荷脑促渗作用较好,但其合用效果小于3%氮酮单用的效果。结论:氮酮单用有较好的促渗效果,即3%氮酮与其他促渗剂合用不利于马钱子碱的体外透皮吸收。
OBJECTIVE:To study the effect of different penetration enhancers on transdermal permeation of brucine patch in vitro.METHODS:Brucine patch was prepared with different concentration and type of penetration enhancers.Using modified Franz diffusion cell and excised male rat skin as model,the concentration of brucine was determined by HPLC.Accumulative permeation quantity and velocity constant of brucine were fitted.RESULTS:3% azone was used to prepare brucine patch which was characterized with sound accumulative permeation velocity and permeation quantity.3% azone combined with 3% menthol promoted the transdermal permeation of brucine patch,while 3% azone alone was the best.CONCLUSION:Brucine patch prepared by 3% azone has higher transdermal rate than others,combination of 3% azone and other penetration enhancers is not good for transdermal absorption of brucine.
