摘要
目的检测MAGE-1、NY-ESO-1、SCP-1基因在直肠癌中的表达情况,探索其在直肠癌免疫治疗中的应用价值。方法应用MAGE-1、NY-ESO-1、SCP-1单克隆抗体,对68例直肠癌组织进行免疫组织化学法(SP法)检测,SPSS统计软件分析所得数据。结果在检测的68例直肠癌标本中,MAGE-1、NY-ESO-1、SCP-1的阳性表达率分别为30.9%(21/68)、23.5%(16/68)、17.6%(12/68)。多个CTA基因可在直肠癌中同时表达,至少表达一种CTA的频率为67.7%(46/68),同时表达两种或两种以上CTA的频率是20.6%(14/68),同时表达三种CTA的频率为5.9%(4/68)。阳性颗粒均位于细胞质。MAGE-1、NY-ESO-1、SCP-1基因的表达主要集中在肿瘤细胞分化较好Ⅰ级直肠腺癌,阳性率有显著性差异(P<0.05),与年龄、性别、肿瘤大小、Dukes分期和淋巴结转移无显著相关性(P>0.05)。结论 CT抗原(MAGE-1、NY-ESO-1、SCP-1)在直肠癌组织中有高特异的表达,这使得用这些CTA编码的蛋白作为疫苗用于直肠癌的免疫治疗成为可能。
Objective To investigate the expression of cancer-testis CT antigens MAGE-1、NY-ESO-1、SCP-1 genes in human colorectal carcinomas and the possibility of applying these antigens as targets for specific immunotherapy for colorectal carcinomas.Methods Using immunohistochemical techniques to analyze the expression of MAGE-1、NY-ESO-1 and SCP-1 in 68 cases of human colorectal carcinomas.Statistial analysis was done with SPSS program.Results Expression of MAGE-1、NY-ESO-1 and SCP-1 was observed in 30.9%(21/68)、23.5%(16/68) and 17.6%(12/68) of colorectal carcinomas samples.46(67.7%) expressed at least one CT antigen,and 14(20.6%)expressed at least two and three CT antigen and 4(5.9%)expressed three CT antigen.Inmunohistochemical staining was only detectable in the cytoplasm.MAGE-1、NY-ESO-1 and SCP-1 expression was found more frequently in well differentiated tumors,statistical significance wsa found between different pathologic grade(p0.05).Expression rate of MAGE-1、NY-ESO-1 and SCP-1 has no relationship,however,with age,gender,tumor size,Dukes and lymph node metastasis(P0.05).Conclusion CT antigens(MAGE-1、NY-ESO-1、SCP-1) are expressed with high specificity in human colorectal carcinomas and their products may provide targets for antigen-specific immunotherapy of human colorectal carcinomas.
