摘要
目的:通过观察脑梗塞大鼠神经功能及S-100β蛋白、一氧化氮(NO)的表达,探讨逐痰通络汤对急性脑梗塞大鼠的影响。方法:SD大鼠72只随机分假手术组、对照组、药物组,各组分术后1天、3天、7天、14天(n=6只)。参照Longa线栓法建立大脑中动脉栓塞(MCAO)模型,在各时间点进行神经功能评分,并留取脑组织免疫组化检测S-100β蛋白,留取血清检测一氧化氮含量。结果:①与假手术组比,对照组、药物组神经功能评分、S-100β及NO明显增高(P<0.01)。②与对照组比,药物组在各时间点神经功能评分低,S-100β蛋白阳性表达在3天、7天明显减少(P<0.05);毒性NO表达较少,其中第3天差异显著(P<0.05),。结论:逐痰通络汤可促进急性脑梗塞大鼠神经功能的恢复,其作用可能与抑制S-100β蛋白、毒性NO的释放有关。
Objective:By observeing the expression of S-100β,the content of nitric oxide(NO) to study the role of Zhutantongluo Recipe in acute focal cerebral infarction of rats.Methods:A total of 72 sprague-Dawley male rats were divided randomly into 3 groups as follows: sham groups,match groups and medicine groups.Each of groups were divided to four groups(First day,third day,seventh day and fifteenth day)with six rats.Set up models with middle cerebral artery occlusion(MCAO)using monofilament.Judge scores of neural functional defect and check The expression of s-100βand the content of NO at the different time.Results:①To compare the sham groups’,they had significant increase at neural functional defect,S-100βand NO(P〈0.01).②To compare the match groups’,medicine groups were lower at neural functional defect,the expression of S-100βon the 3rd day and 7th day(P〈0.05)and nitric oxide on the 3rd day(P〈0.05).Conclusion:Zhutantongluo Recipe may improve the recovery of neural function.Its Protective mechanism may relate to release the overexp-ression of S-100βproteinum and iNOS.
