山柰酚标准品的制备及含量测定

摘要 以天然提取的山奈酚粗品为原料,对其进行纯化工艺的研究以获得山柰酚标准品。采用冰醋酸作为结晶溶剂,对山柰酚进行纯化精制,并采用HPLC方法对其进行含量测定,以Kromasil KR100-5C18(250mm×4.6mm,5μm)为色谱柱,乙腈-0.1%磷酸溶液(35:65)为流动相,流速1.0mL μmin^-1,柱温30℃,检测波长366nm。采用面积归一化法测得产品含量为99.70%,收率为67%。山柰酚的线性范围在5—500μg·mL^-1(r=0.9997)。该方法操作简单,实用性强,为大规模制备山奈酚标准品提供了参考方法。 To obtain standad sample of kempferol from natural extracted sample, acetic acid was used as reerystalization. A kromasil KR100-5 C18 column(was 250 mm×4.6 mm,5 μm) used with a mixture of acetonitrile and 0. 1% phosphoric acid solution (35:65) as the mobile phase at a flow rate of 1.0 mL/min,the column temperature was 30 ℃ with the detection wavelength at 366 nm. The purification yield was above above 67%, and the content was up to 99.70%. The linear curve was obtained in the range of 5 - 500μg·mL^-1( r = 0. 9997 ). This method is simple, practical, and it provides a way to prepare high purity kaempferol for industry.
