摘要
目的:建立测定双黄连注射液中咖啡酸含量的高效液相色谱法。方法:采Agilent XDB-C18色谱柱(4.6mm×150mm×5μm),流动相以甲醇为流动相A,以0.5%磷酸溶液为流动相B,进行梯度洗脱;进样量5μl;流速1ml/min;检测波长为324nm。结果:咖啡酸在进样量在0.03~0.30μg范围内与峰面积线性关系良好(r=0.9997),平均回收率为99.42%(RSD=1.18%)。结论:方法专属性强、准确、快速,适用于双黄连注射液的质量控制。
Objective:To establish a RP-HPLC method for the determination of coffeic acid in Shuanghuanglian Injection.Methods:The column was Agilent XDB-C18(4.6 mm×150 mm×5 μm).The mobile phase consisted of methanol-0.5% phosphoric acid with gradient elution;the inject volumes was 5 μl;theflow rate was 1.0 mL/min and the detection wavelength was at 324 nm.Results:The calibration curves were linear within the range of 0.03-0.30 μg(r=0.999 7) for coffeic acid.The average recovery was 99.42%(RSD=1.18%).Conclusion:The method is rapid and accurate.It can be used for quality control of Shuanghuanglian Injection.
