摘要
光作用于结晶溶液体系促使处于亚稳区的蛋白质结晶的方法是近年来新发展的一种技术。由于该技术有着重要的潜在应用价值,引起了人们的关注。至今,人们已针对该技术做了大量的研究工作,并取得了一系列重要进展,研究发现利用大功率氙灯光源短时间照蛋白质结晶溶液体系会发生光化学反应,并最终促使蛋白质结晶;而利用脉冲激光短时间照射蛋白质结晶溶液体系,由于激光的空穴效应也可以促使蛋白质结晶。本文从光对结晶溶液体系产生的效应入手,评述了光照诱导蛋白质结晶及其机理的研究进展,并展望了该技术的未来发展前景。
Light-induced crystallization of protein in metastable solution is a new technique developed in recent years.This technique has attracted much attention for its potential application in practical protein crystallization.So far,significant progresses have been made in protein crystallization induced by using light irradiation.It was found that short irradiation of high-power Xe lamp on the crystallization solution of protein can trigger photo-chemical reaction and further induce the nucleation.It was also found that pulsed laser irradiation of the crystallization solution of protein will induce the nucleation of crystals due to the cavity bubble generation caused by laser irradiation.In this paper,the effect of light-induced protein crystallization and its mechanism are reviewed and discussed,and the future prospect of this technique is proposed.
