摘要
目的:检测Ag85A DNA疫苗在小鼠肠道M细胞的表达,探索口服疫苗在肠道局部的摄取方式。方法:将C57BL/6小鼠随机分为2组,即生理盐水组和脂质体包裹重组质粒组。分别将生理盐水和脂质体包裹pCDNA3.1+/Ag85A以灌胃方式投给各组小鼠,共免疫3次,每次间隔14 d,末次免疫后14 d处死小鼠,取肠,用免疫荧光方法检测Ag85A在肠道局部M细胞的表达。结果:Ag85A DNA疫苗在小鼠肠道局部M细胞有表达。结论:口服脂质体包裹的DNA疫苗可以被肠道M细胞摄取。
Objective:To explore the intake of oral DNA vaccine.Method:C57BL/6 mice were randomly divided into two groups,PBS control and the recombinant plasmid parceled with liposome,respectively intragastric administration.The mice were immunized every 14 days,three times altogether.After another 14 days,the expression of Ag85A in intestinal M cells was detected with immunofluorescence.Results:Ag85A recombinant DNA vaccines were expressed in mouse intestinal M cells.Conclusion:Oral DNA vaccine parceled with liposome can be intaken by intestinal M cells.
