摘要
采用响应面分析法优化短小芽孢杆菌木聚糖酶在PEG2000/K2HPO4双水相体系中的萃取条件。确定最佳萃取条件为NaCl 6%、PEG 2000 18%、K2HPO4 10%。在最佳萃取条件下得木聚糖酶分配系数为3.38,蛋白分配系数为0.13,纯化倍数为7.93,回收率为84.24%。考察了萃取酶液的部分酶学性质,其最适温度为50℃,最适pH为7.0。
The operating condition of aqueous two-phase systems(ATPS) was optimized to extract xylanase from Bacillus pumilus,using response surface analysis.The optimal system contained NaCl 6 %,PEG2000 18 % and K2HPO4 10 %,respectively.Resulting in a portion factor of xylanase and protein 3.38,0.13,respectively,a purification factor of 7.93 and a 84.24 % yield of enzyme activity.The characteristics of the partially purified xylanase were also studied and results showed that the optimal temperature and pH of it was 50 ℃ and 7.0,respectively.
