摘要
目的探讨急性胰腺炎(acute pancreatitis,AP)大鼠胰腺组织一氧化氮(nitric oxide,NO)的表达及肠壁通透性的改变,并观察瘦素(Leptin)对其的影响。方法大鼠随机分为3组,①AP组(n=15);②治疗组(n=15);③假手术对照组(n=15)。测定胰腺组织中NO的表达,血淀粉酶(blood amylase,AMY)与肠内^(125)I-清蛋白累积指数,以及胰腺和回肠病理检查。结果 AP组胰腺NO呈强阳性表达,血中AMY及^(125)I-清蛋白累积指数和腹水量均显著高于假手术对照组(P<0.05)。与AP组比较,治疗组胰腺及回肠病理损害明显减轻,胰腺NO表达强度较弱,血中AMY及腹水量均显著下降(P<0.05),^(125)I-清蛋白累积指数亦降低。结论 AP肠壁通透性增加的原因之一,可能与AP时胰腺产生过多NO有关。Leptin可以使AP时胰腺NO的表达及肠壁通透性明显降低,对AP有一定预防治疗作用。
Objective To investigate the expression of NO and change of gut permeability and the effect of Leptin on them in rats with acute pancreatitis (AP). Methods All rats were randomly divided into three groups : Sham operation( n = 15), AP group( n = 15), and Leptin therapeutic group ( n = 15). The NO expression of pancreas, blood amylase(AMY) and 125I - labeled serum albumin were examined. The histopathologic changes of pancreas and ileum were observed. Results The NO expression of pancreas in AP group were markedly increased, The blood AMY and intestinal permeability in AP group were much higher than those of sham operation group ( P 〈 0.05 ). While the NO expression of pancreas, the blood AMY and intestinal permeability in Leptin therapeutic group decreased markedly in comparison with AP group. The pathological changes of pancreas and ileum in group AP were more severe than in Leptin therapeutic group ( P 〈 0.05 ) . Conclusion Excess NO production may be a factor related to intestinal permeability in AP. Leptin can down - regulate the NO expression of pancreas and depress the intestinal permeability in AP.
