摘要
目的通过测定兔血小板聚集和TXB2的含量,比较观察F、I、FI、FIS在抗血栓方面的药理作用特点。方法用比浊法[4]和放免法[7]测定F、I、FI、FIS在体外对兔血小板聚集及释放的TXB2的影响。结果连有阿魏酸和吲哚美辛的载体衍生物FI,FIS可抑制AA、ADP诱导的血小板聚集,在10~250μmol·L-1的剂量范围内,两药的作用呈剂量依赖关系,FI、FIS240μmol·L-1对AA和ADP诱导的兔血小板聚集的抑制率分别为59.32%、58.58%和39.09%、49.56%。FI、FIS240μmol·L-1对TXB2产生的抑制率分别为40.60%、28.72%。结论在等分子浓度下,FI、FIS对AA诱导的兔血小板聚集及其TXB2和释放的抑制作用明显强于阿魏酸和吲哚美辛本身。
Objective To investigate the effect of
IndomethacinumFerulate acid(FI) and IndomethacinumFerulate acidstarch (FIS) on rabbit platelet
aggregation induced by ADP and AA and thromboxane B2production. Methods Influences of
F,I,FI,FIS on rabbit platelet aggregation and TXB2 production were assaied by born GVR and
RIA. Results Both FI and FIS 12.5,60.0,240.0 molL-1,could inhibit the rabbits platelets
aggregation induced by adenosin diphosphate (ADP,2.0molL-1),arachidonic acid (AA,33.3
molL-1).Which showed a good doseeffect relationship.The inhibitions by 10300 molL-1 were
dependent FI 25.059.32%,FIS 13.1239.09%(ADPinduced),FI 29.96%63.06%),FIS
11.47%39.56%(AAinduced).inhibit the ptoduction of TXB2 as well,(AAinduced)FI 40.60%,FIS
28.74%(240.0 molL-1)for TXB2. Conclusions There are positive correlations between the
inhibition of platelet aggregation and TXB2 and effect mechanism on speranskia tuberculata.
