灯盏花素对糖尿病大鼠肾脏PI3K/AKT传导通路的影响 被引量:12

摘要 目的探讨灯盏花素对糖尿病大鼠肾脏PI3K/AKT传导通路的影响.方法 80只健康昆明种雄性SD大鼠随机分为正常对照组(N)、糖尿病组(DM)各25只和糖尿病灯盏花治疗组(EB)30只.链脲菌素(streptozotocin,STZ)诱导建立糖尿病大鼠模型.EB组灯盏花素20 mg/(kg.d)腹腔内注射,给药后2周和6周处死大鼠,SYBRGreenI荧光定量RT-PCR方法检测各组大鼠肾脏AKT、bax和bcl-2 mRNA的表达.检测血肌酐、尿素氮和血糖水平;肾组织HE,PAS染色比较各组肾小球平均面积(MGPA)、肾小球平均体积(MGV).结果 (1)肾脏指数、MGPA、MGV为糖尿病组>灯盏花组>正常组(P<0.05);(2)血肌酐和尿素氮水平为糖尿病组>灯盏花组>正常组(P<0.01).(3)与正常组比较糖尿病组大鼠肾脏bax、bcl-2 mRNA的表达显著增强,bax/bcl-2的比值明显升高;灯盏花组与糖尿病组比较bcl-2 mRNA表达增强,bax mRNA表达减弱,bax/bcl-2比值降低.AKTmRNA表达,2周各组间无差异,6周EB组较N组增高(P<0.01),较DM及2周EB组增高(P<0.05).结论高血糖可促进糖尿病大鼠肾脏肥大,影响肾功能;灯盏花素可增加糖尿病大鼠肾脏AKTmRNA的表达,进而增加抗凋亡基因bcl-2的表达,影响NF-κB的表达,抑制糖尿病大鼠肾脏肥大,发挥保护肾功能. Objective To investigate the effect of Erigeron breviscapine on the conduction pathway of PI3K/AKT in kidney of diabetic rats.Methods Eighty healthy male rats were selected and randomly divided into three groups:25 in normal group(N),25 in diabetic group(DM),and 30 in breviscapine treatment group(EB).Diabetic rats in DM and EB group were induced by injection of streptozotocin(STZ).Erigeron breviscapine was injected intraperitoneally into rats in EB group 20 mg/(kg.d).Rats in three groups were killed after 2 weeks and 6 weeks of giving medicine.Expression of AKT,bax and bcl-2 mRNA in the kidney were detected by real-time quantitative RT-PCRwith SYBR Green I.The blood samples were collected to examine creatinine(Cre),urea nitrogen(Bun)and glucose.Rats' renal tissues were also collected to compare MGPA,MGV among the three groups by the HE and PAS staining.Results(1)The kidney index,MGPA,and MGV of rats were in DM groupEB groupN group(P0.05).(2)Serum levels of Cre and Bun of rats were in DM groupEB groupN group(P0.01).(3)Compared with N group,the expression of bax and bcl-2 mRNA in DM group was increased obviously,the ratio of bax/bcl-2 was also increased.Compared with DM group,the expression of bcl-2 mRNA in EB group was increased,while bax mRNA was decreased,the ratio of bax/bcl-2 was decreased.The expression of AKT mRNA in two weeks had no significant difference among the three groups,but was increased in 6 weeks in EB group,compared with that in N group(P0.01),DM and the 2 week's EB group(P0.05).Conclusions In diabetic rats,the hyperglycosemia may aggravate the glomerulopathy and affect the renal function.Breviscapine has renal protection on diabetic nephropathy through increasing the expression of AKT mRNA,increasing the expression of bcl-2 mRNA,affecting the expression of NF-KB,and inhibiting kidney hypertrophy of diabetic rats.
