摘要
目的探讨卵泡刺激素受体(folicle-stimulating hormone receptor,FSHR)基因多态性与不孕妇女卵巢反应的关系。方法提取不孕妇女外周血白细胞基因组DNA,用PCR法扩增FSHR基因外显子10的片段并测序,研究其基因多态性。结果①120例不孕妇女中FSHR在基因外显子10的680多态位点上有3种基因型,分别为Asn680Asn(A/A)型占60%、Asn680Ser(A/S)型占17.5%、Ser680Ser(S/S)型占22.5%。②不同卵巢反应患者的基因型构成比为:卵巢低反应组中A/S型者占29.41%,A/S型者占11.76%,S/S型者占58.82%;卵巢正常反应组中A/A型者占62.65%,A/S型者占19.28%,S/S型者占18.07%;卵巢高反应组中A/A型者占75%,A/S型者占15%,S/S型者占10%。③A/A型与S/S型的卵巢反应性相比较:P<0.05,差异有统计学意义。结论①不孕妇女FSHR外显子10的680位存在A/A、A/S、S/S 3种基因型。②3种基因型中S/S型在卵巢低反应妇女中出现频率较高,可能与不孕妇女卵巢低反应有关。A/A型在卵巢高反应和正常反应妇女中出现频率较高,可能与不孕妇女卵巢正常反应或高反应有关。
Objective To investigate the relationship between follicle-stimulating hormone receptor(FSHR) polymorphisms and ovarian response in infertile women.Methods DNA was extracted from leukocytes from the peripheral blood of 120 infertile women.Exon 10 of the FSHR was amplified by PCR and the DNA fragments were sequenced to determine the genotype of FSHR.Results Among the 120 infertile women,Asn680Asn(A/A) was present in 72(60%) women,Asn680Ser(A/S) in 21(17.5%) women,and Ser680Ser(S/S) in 27(22.5%) women.The distribution of three genotypes was 29.41% for A/A,11.76% for A/S and 58.82% for S/S in women with poor ovarian response,respectively.The distribution was 62.65% for A/A,19.28% for A/S and 18.07% for S/S in women with normal ovarian response,respectively.The distribution was 75% for A/A,15% for A/S and 10% for S/S in women with high ovarian response,respectively.The ovarian response to gonadotropin in genotype S/S was lower than that in genotype A/A(P0.05).Conclusion There are three FSHR genotypes(A/A,A/S and S/S) at position 680 in exon 10 of infertile women.Genotype S/S predominates in infertile women with poor ovarian response and A/A predominates in infertile women with high ovarian response or normal ovarian response,suggesting that S/S is related to poor ovarian response and A/A is related to ovarian sensitivity.
