摘要
目的建立高效液相-质谱联用(LC/MS/MS)法测定人血浆盐酸伪麻黄碱浓度的方法。方法采用Thermo Hypurity C18色谱柱(150mm×2.1mm,5μm);流动相:5mmol.L-1醋酸铵-甲醇(70:30);流速:0.3mL.min-1;柱温:40℃;MRM模式检测(离子对),内标法定量。结果血浆盐酸伪麻黄碱在17.67~565.30ng.mL-1浓度范围内线性关系良好(r=0.9995)。低、中、高3种浓度相对回收率分别为101.96%,102.08%,98.85%,日内、日间RSD均〈10%。结论该方法快速简便、灵敏准确,可用于盐酸伪麻黄碱血浆浓度的测定及其药动学研究。
Objective To establish a LC/MS/MS method for determination of pseudoephedrine hydrochloride in human plasma.Methods The analysis was carried out by using Thermo Hypurity C18 column(150 mm× 2.1 mm,5 μm).The mobile phase consisted of 5 mmol·L-1 ammonium acetate-methanol(70:30)with a flow rate at 0.3 mL·min-1.The column temperature was set at 40 ℃ and concentrations in plasma were determined at ion pairs(166.2/148.2)by MRM of LC/MS/MS.Results The linear range of pseudoephedrine hydrochloride was 17.67~565.3 ng·mL-1(r=0.999 5),the recoveries were 101.96%,102.08%,98.85%,at low,middle and high concentrations,respectively,and RSD were below 10%.Conclusion The method is simple and precise,which can be used for determination of pseudoephedrine hydrochloride in human plasma and its pharmacokinetics study.
