摘要 [目的]建立鹿药Smilacina japonica A.Gray中活性成分——黄酮类化合物含量的测定方法。[方法]采用铝盐络合显色分光光度法,以芦丁为对照品,NaNO2-Al(NO3)3-NaOH为显色剂,采用正交试验确定显色剂各组分的最佳用量,在505 nm波长处测定光密度,计算鹿药中的总黄酮含量。[结果]正交试验结果表明,最佳显色剂各组分用量为50 mg/ml NaNO22.0 ml,100 mg/ml Al(NO3)30.5 ml,40mg/ml NaOH15.0 ml。该测定方法在浓度为0.004-0.028 mg/ml时,芦丁光密度与浓度的线性关系良好,R^2=0.999 6。鹿药植株不同部位总黄酮的含量,以鹿药茎叶中较高,为1.8%,根茎中为0.43%,种子中含量极低;不同采收时期中,以9月中旬采收的根茎中总黄酮含量较高。[结论]该方法操作简便、快速、可行,可用于测定鹿药不同部位及不同采收期根茎中总黄酮含量,研究结果为鹿药的质量评价及开发利用提供了理论依据。 [Objective] To establish a method for determining total flavonoids in Smilacina japonica A.Gray.[Method]Taking rutin as control,the total flavonoids were determined by ultraviolet-visible spectrophotometry at 505 nm,the color reagent was a mixture of NaNO2-Al(NO3)3-NaOH solution,the best color reagent formula was determined by the orthogonal experiment.[Result] The orthogonal experiment results showed that the best color reagent formula was 2.0 ml 50 mg/ml NaNO2,0.5 ml 100 mg/ml Al(NO3)3 and 15.0 ml 40 mg/ml NaOH.There was a good linear relationship between the total flavonoids content and absorbance in the concentration range of 0.004-0.028 mg/ml,R2=0.999 6.Among the different parts,the contents of total flavonoids in the stem leaf was higher,reached 1.8%;in the rhizome was lower,only 0.43%;the lowest in the seed.The contents of total flavonoids in the rhizome of Smilacina japonica at the middle ten days of September was the highest among the different harvesting time.[Conclusion] The method was simple,fast and accurate,and it had been applied to determine flavonoids in Smilacina japonica successfully;these primary research results supplied credible foundation for quality estimate and utilization of Smilacina japonica A.Gray.
