摘要
[目的]提取高质量的山药叶片总RNA。[方法]以山药叶片为材料,采用异硫氰酸胍法、改良CTAB法提取山药叶片总RNA并进行比较分析,利用琼脂糖凝胶电泳分析所提RNA的完整性,并用紫外分光光度计分析RNA的纯度。[结果]改良CTAB法提取的总RNA的28S、18SrRNA条带清晰、整齐,测得A260/A280为1.72.0。[结论]改良的CTAB法适合山药叶片总RNA提取。
[Objective]The aim was to obtain high-quality total RNA from leaves of Dioscorea opposita.[Method]The guanidinium isothiocyanate method and modified CTAB method was used to extract total RNA from leaves of Dioscorea opposita respectively.By employing agarose gel electrophoresis and UV-spectrophotometer,the integrity and purity of total RNA were analyzed.[Result]The 28S and 18S bands of total RNA extracted by the modified CTAB method were clear and visible,and the A260/A280 value of it was from 1.7 to 2.0.[Conclusion]Modified CTAB method was suitable for the extraction of total RNA from Dioscorea opposita leaves.
