乙酰水杨酸对某些细胞因子的影响与胃溃疡复发 被引量:1

摘要 目的探讨胃溃疡复发的机制。方法用乙酰水杨酸诱导已愈合的溃疡复发。在溃疡复制后的第55天用免疫组化方法检测表皮生长因子、血管内皮生长因子和核因子-κB的表达,用酶联免疫吸附测定法测定了肿瘤坏死因子-α的含量,还检测了黏膜下炎细胞数量。结果乙酰水杨酸组的表皮生长因子和血管内皮生长因子的积分光密度低于模型对照组和盐水对照组(P<0.01);乙酰水杨酸组的核因子-κB积分光密度,肿瘤坏死因子-α的含量和黏膜下炎细胞数量高于模型对照组和盐水对照组(P<0.01)。结论乙酰水杨酸可能抑制表皮生长因子、血管内皮生长因子的表达;通过增加核因子-κB的表达促进肿瘤坏死因子-α生成增多,导致黏膜下炎细胞数增加,引起溃疡复发。 Objective To explore the mechanisms of gastric ulcer recurrence. Methods With aeetylsalieylie acid, recurrence of healed ulcer was induced. On the 55th day after ulcer induction, the expressions of the epidermal growth factor, the vascular endothelial growth factor and the nuclear factor - KB were observed by immunohistoehemistry method. The contents of the tumor necrosis factor - α were determined with enzyme linked immunosorbent assay. The number of inflammatory cells in submucous tissue was observed. Results The integral light density of epidermal growth factor and the vascular endothelial growth factor were decreased in acetylsalieylic acid group as compared with those of model or saline control groups (P 〈 0.01 ). The integral light density of the nuclear factor - KB, the contents of tumor necrosis factor - c~ and the number of the inflammatory cells in submucous tissue were increased in acetylsalieylic acid group as com- pared with those of model or saline control groups (P 〈 0.01 ). Conclusion Acetylsalicylic acid could inhibit the expression of the epidermal growth factor and the vascular endothelial growth factor, and promote the production of the tumor necrosis factor - α by increasing the expression of the nuclear factor - KB, which will lead to the increasing of the number of the inflammatory cells in submucous tissue. These will result in the ulcer recurrence.
