摘要
对几种不同的质粒在相同宿主中的稳定性进行检测,结果表明,在非选择性压力下,4种不同的质粒在大肠杆菌宿主细胞中的稳定性有明显的差异,其中pUCG418最稳定,该质粒失去的速率仅为0.5%;pEMBLYe23R次之;pUC9很不稳定,在群体细胞中,无质粒细胞高达65.8%;而质粒pG418最不稳定,当细胞增殖到29代时,细胞丢失该质粒的比例高达100%.DNA琼脂糖凝胶电泳的结果表明,这几种质粒的不稳定为分离不稳定性.
The stabilities of several plasmids in Escherichia coli host cells were determined. The result showed remarkable defference in stability from one another. One of them,PUC G418 was the most stable one,the plasmid had been lost only by 0.5%,and pEMBL Ye 23R was stable, while pUC 9 was very instable,the plasmid -free cells, took up 65. 8% of the population of the hod cells. Plasmid pGBTe G418 wasthe most instable one in E. coli JM101, the cells, of poputation had lost completely the capa bility of resistence to ampicilin when the cells reached the 29th generation. DNA agarose gel electrophoresis indicated that instability f these plasmids was called segregation instablity.
