九龙牦牛IFN-tau基因的克隆测序及聚类分析

摘要 研究旨在分析九龙牦牛的干扰素-τ(IFN-tau)基因与其他属种的差异.利用PCR对九龙牦牛的IFN-tau进行克隆并测序,并将该基因的氨基酸序列与其他属种进行聚类分析.结果表明:利用基因组DNA扩增得到的九龙牦牛IFN-tau基因为588bp,与普通牛有98.25%以上的相似度.通过系统发育分析,大额牛、亚洲水牛为一支,山羊、绵羊为一支,美洲野牛、欧洲牛、九龙牦牛为一支,林麝单独为一支. The objective of this study is to identify the differences of interferon-tau(IFN-tau) between Jiulong Yak and other Genus.IFN-tau of Jiulong yak is cloned and sequenced.The predicted amino-acid sequence is compared with other Genus using cluster analysis software.The result shows that a 588bp Jiulong yak IFN-tau gene is amplified using genomic DNA as the template.The Jiulong yak IFN-tau gene is 98.25% similar to the homologous gene of Bos taurus.Phylograms constructed on the basis of INHα mature peptide sequences from various species show that mithun and buffalo comprise one clade,and goat and sheep comprise another.Bison,cattle and Jiulong yak comprise a third clade,where a moschus berezosvskii represents an entirely different clade.
