摘要
目的:探讨汉防己碱诱导乳腺癌细胞MCF-7凋亡的信号机制。方法:以MTT法测定不同浓度的汉防己碱(1、5和10μmol/L)对MCF-7肿瘤细胞增殖抑制;流式细胞仪检测细胞凋亡;免疫印迹法分析AKT活化;细胞免疫化学染色检测NF-κB表达。结果:不同浓度汉防己碱对培养的乳腺癌细胞MCF-7生长有显著的抑制作用,有效的诱导凋亡,且呈剂量依赖性,各组MCF-7细胞生长抑制率分别为(34.04±2.06)%、(54.07±1.08)%和(70.29±1.05)%,与对照相比较差异均有统计学意义,P<0.01;各组细胞凋亡率分别为(15.21±2.13)%、(23.07±1.08)%和(30.35±1.06)%,与对照相比差异均有统计学意义,P<0.05。汉防己碱对细胞内总AKT激酶蛋白的表达没有影响,但抑制磷酸化AKT蛋白的表达;同时对细胞内NF-κB的表达有抑制作用。结论:汉防己碱诱导乳腺癌细胞凋亡的机制与Akt/NF-κB信号途径有关。
OBJECTIVE:To study the cell signal mechanism of tetrandrine on apoptosis of human breast cancer cell line MCF-7. METHODS: The effects of different doses (1, 5 and 10 μmol/L) of tetrandrine on the proliferation of MCF-7cells were observed by MTT assay. Apoptosis was detected with flow cytometry. Western blot was performed to assess ser473-phosphorylaed Akt. Immuno-cytochemistry was used to analyze the expression of NF-κB. RESULTS: The different doses of tetrandrine inhibited the proliferation and induced the apoptosis of MCF-7 in a dose-dependent manner, and the inhibitory rates of proliferation in MCF-7 cells were (34.04±2.06)%,(54.07±1.08)% and (70.29±1.05)% respectively (P0.01); the rates of apoptosis were (15.21±2.13)%, (23.07%±1.08)% and (30.35±1.06)% respectively (P0.05). Tetrandrine could significantly inhibite serine 473 phosphorylation in Akt and the expression of NF-κB. CONCLUSION: Tetrandrine can induce apoptosis in MCF-7 cells through the Akt/NF-κB signal pathway.
