一种改进的人骨髓间充质干细胞培养方法

摘要 目的提高体外培养人骨髓间充质干细胞(HMSCs)的获得率和扩增率。方法在无菌条件下采用骨髓穿刺术提取健康人骨髓2~3ml加入含10%胎牛血清(FBS)的α-MEM培养基中,按2×106/cm2密度接种于培养瓶中,置于培养箱中培养。第3天半量换液,第6天用PBS冲洗三遍后全量换液,待细胞生长至80%~90%融合时,消化传代;倒置显微镜进行细胞形态学观察;MTT法测定细胞P1和P5生长曲线;应用流式细胞仪检测细胞表面CD34和CD29抗原的表达进行细胞鉴定。结果分离HMCs大小较为均匀,基本上呈梭形或星形;细胞生长曲线测定表明接种后第3天细胞进入指数增生期,至第5天进入平台期,随着传代次数的增加,细胞增殖速度变慢;经流式细胞仪检测显示CD34阴性、CD29阳性。结论本实验采用贴壁法培养的骨髓细胞为HMSCs,本培养体系能有效地提高HMSCs的获得率和扩增率。
