摘要
建立了一种采用高效液相色谱快速检测2-氯-6-甲基苯胺含量的方法。色谱柱为shim-pack Vp-ODS C18柱(150mm×4.6mm),紫外检测器,检测波长238nm,柱温25℃,流动相为V(甲醇):V(水)=80:20,流动相流速1.5mL/min,2-氯-6-甲基苯胺的含量在0.461~2.304mg/mL范围内与色谱峰面积线性关系良好,相关系数为0.9996,该方法测定结果的相对标准偏差为0.22%(n=6),及加样回收率为99.49%。利用本法测定出实验室合成粗品中2-氯-6-甲基苯胺含量为49.05%。
A high performance liquid chromatography method is devised for detecting 2-chloro-6-methylaniline.Chromatographic separation is performed on the shim-pack Vp-ODS C18 column(150mm×4.6mm) at 25℃,with an eluant of methanol: water=80:20(volume)flowing at a rate of 1.5mL/min.The eluate is monitored by the UV detector,the detecting wavelength being 238nm.The linear range of 2-chloro-6-methylaniline is 0.461~2.304mg/mL(r=0.9996).The relative standard tolerance of results detected by this method is 0.22%(n=6) and the average recovery is 99.49%.It is shown that,by using this method,of the crude product synthesized in the lab,the net content of 2-chloro-6-methylaniline detected reaches 49.05%.
