摘要
目的:研究肠易激综合征(Irritable bowel syndrome,IBS)模型大鼠结肠组织5-羟色胺(5-hydroxytryptamine,5-HT)含量、5-HT转化率以及眼针治疗的影响。方法:实验动物分成对照组、IBS模型组、眼针组,采用束缚刺激与应激刺激相结合的方法建立IBS动物模型并进行眼针治疗。各组采用ELISA法检测结肠组织5-HT和5-羟吲哚乙酸(5-hydroxyindoleacetic acid,5-HIAA)的表达,采用RT-PCR法检测结肠组织5-HT重摄取转运体(serotonin reuptaketransporter,SERT)、色氨酸羟化酶1(Tryptophan hydroxylase1,TPH1)的mRNA表达。结果:与对照组比较,IBS模型组5-HT、5-HIAA水平显著升高,5-HT转化率也显著升高,SERT mRNA表达下调而TPH1mRNA表达上调;与IBS模型组比较,眼针组5-HT和5-HIAA水平明显下降,5-HT转化率显著降低,SERT mRNA表达上调而TPH1mRNA表达下调。结论:5-HT降解与合成分泌的不相匹配可能是IBS模型大鼠5-HT水平升高的重要原因,眼针治疗可有效改善IBS大鼠5-HT代谢,使5-HT恢复至正常水平。
Objective:To investigate colonic 5-HT content and percent conversion in the rat with IBS and effects of ocular acupuncture therapy( OAT). Methods:Animals were classified into control??IBS model and OAT groups. Rat model of IBS was established by method of stress and combining bound,and OAT was given. Colonic 5 -HT and 5 -HIAA of each group were examined by ELISA. SERTmRNA and TPH1mRNA were examined by RT -PCR. Results:Compared with those in control,the levels of 5-HT and 5-HIAA increased,percent conversion of 5-HT increased too,expression of SERTmRNA decreased and TPH1mRNA expression increased in IBS model. Compared with those in IBS model,the levels of 5 -HT and 5 -HIAA and percent conversion of 5-HT decreased,expression of SERTmRNA increased and TPH1mRNA expression decreased in OAT. Conclusion:The difference of 5 -HT between anabolic metabolism and catabolism in IBS model suggestes that the disharmony is involved in the pathogenesis of IBS. OAT can correct 5-HT metabolism effectively.
