几种土壤微生物总DNA提取方法的比较 被引量:3

Comparison of Several Methods for the Extraction of Total Community DNA from Soil

摘要 采用Bourrain法、Beadbeating法和Martin-Laurent法提取土壤微生物总DNA,并以细菌16SrD-NA通用引物进行PCR扩增。结果表明,3种方法提取的DNA大小都在23.1kb以上。Bourrain法提取到的土壤微生物总DNA量最多,但是蛋白质和腐植酸含量等杂质含量也最高;而Martin-Laurent法提取的DNA量小于Bourrain法,并且含有较多的杂质;Beadbeating法提取的土壤微生物总DNA量较Bourrain法少,腐植酸及蛋白质等杂质含量最少。 The total community DNA from soil was extracted by the Bourrain method,Martin-Laurent method and Bead beating method respectively,then the universal primers for 16S rDNA was used for PCR amplification.The results showed that the DNA extractions with different methods were above 23.1kb.Highest amount of DNA and impurities were extracted by Bourrain method compared with other methods.Higher extracts was obtained by Martin-Laurent method.However,lesser pure DNA and little impurities were extracted by Bourrain method.
