摘要
为研究猪链球菌2型在感染猪的组织定位和动态分布,本实验采用30头SPF巴马小型猪,随机分为实验组和对照组。实验组20头,按照1×107CFU·kg-1的剂量肌肉注射ZY05719菌液,对照组10头,按照相同剂量肌肉注射无菌培养基。分别于攻毒后的12h、1d、2d、3d、4d、5d、6d、7d、8d、10d时处死,剖杀并采集样品(扁桃体、淋巴结、肝脏、脾脏、肺脏、肾、脑),制作冰冻切片,并用特异性免疫荧光抗体染色法和细菌分离方法,观察10日内细菌在各组织内的定位和动态分布情况。结果表明,细菌主要定殖于扁桃体的隐窝附近;在脾的边缘区和动脉周围淋巴鞘附近、淋巴结的生发中心和皮质内、肝血窦、肺间质和肾小体中均可见菌体状荧光颗粒;在脑组织中发生脑膜炎的区域,如皮质层等可检出细菌。细菌在肝、肾、肺和脾等实质器官内呈现相似的动力学,感染后的前3d持续上升,第6天开始下降;在扁桃体内12h就能被检出并持续存在;在脑组织中呈现不规律分布。实验结果为研究猪链球菌2型的致病机理提供一定依据。
[Objective] Location and Dynamic distribution of Serotype 2 Streptococcus Suis in Tissues and Organs of Infected SPF Pigs. [Methods] 30 two-month-old SPF pigs were randomly divided into experiment group (include 20) injected with Sichuan strain ZY05719 and control group (include 10) injected with asepsis medium. The SPF pigs were dissected in 12 h, 1 d, 2 d, 3 d, 4 d, 5 d, 6 d, 7 d, 8 d, 10 d after injection. Collect the samples of tonsil, lymphoid node, liver, spleen, lung, brain and make frozen sections, followed by the observations of the localization, and dynamic distribution of SS2 in vivo using immunofluorescence and bacteria isola- tion. [Results] The study indicated that positive S. suis mainly located in tonsil crypts; the fluorescent cell-like particles also can be seen in the marginal zone and periarterial lymphoid sheath of spleen, the germinal center and cortex of lymph node, the hepatic sinu- soid of liver, interstitium of lung; and the bacteria also presented in meningitis region of brain. Dynamics of bacteria in parenchyma- tous organs such as liver, kidney, lung and spleen is similar. The number of bacteria had been persistently rising in 3 days after infection and declined after the 6th day; SS2 could be detected in the tonsil in 12 h after injection and persisted; but the distribution in brain is irregular. [Conclusion] This study can be taken as basic research of SS2 pathogenic mechanism.
