摘要
以苦瓜子叶、上、下胚轴为外植体,在添加不同浓度激素组合的培养基上诱导和继代愈伤组织,比较愈伤组织的状态与生长情况,并对悬浮细胞培养条件进行了优化。结果表明,上下胚轴在添加2,4-D2.0mg·L-1+6-BA4.0mg·L-1或NAA1.0mg·L-1+6-BA4.0mg·L-1的MS培养基上均可诱导出状态良好的愈伤组织,愈伤组织在添加2,4-D0.5mg·L-1+6-BA1.0mg·L-1的继代培养基上能长势最好,添加30g·L-1蔗糖和3.0g·L-1活性炭的B5培养基有利于苦瓜悬浮细胞的快速增殖。
Callus induction and subculture were investigated using cotyledon, epicotyls and hypocotyls as explants in medium including different hormones. Callus quality and growth rate were observed, and condition of cell suspension culture was optimized, too. The results showed that callus should be obtained from epicotyls and hypocotyls, when it adds in MS culture medium 2,4-D 2.0 mg·L-1+6-BA 4.0 mg·L-1 or NAA 1.0 mg·L-1+6-BA 4.0 mg·L-1. The best hormones concentration for callus subculture is 0.5 mg·L-1 2,4-D combination with 1.0 mg·L-1 6-BA. Momordica charantia L suspensional cells proliferate fastly in B5 basal medium supplemented with 30 g·L-1 sugar and 3.0 g·L-1 activated carbon.
