摘要
利用H2O2易通过细胞膜而到达核这一特点,初步探讨了不同浓度H2O2对HL60细胞DNA的氧化损伤程度.发现H2O2浓度在04mmol/L以上时,作用8~24h可以用气相色谱/火焰离子检测器(GC/FID)检测到氧化损伤标志产物———8羟基鸟嘌呤(8ohG),并观测到在04~08mmol/LH2O2作用一定时间时。
Although hydrogen peroxide is generated in cells by normal metabolism, the reaction of the stress or by treatment of the cells with 0 4 mmol/L of hydrogen peroxide for 8~24 hours can result in DNA damage. 8 hydroxyguanine is a typical hydroxyl radical induced products in DNA.The yields of the 8 hydroxyguanine in DNA isolated from H 2O 2 treated HL 60 cells in different ways were reported.The 8 hydroxyguanine in the cells treated in more than 0 4 mmol/L H 2O 2 for eight hours.In order to identify the 8 hydroxyguanine, the result was confirmed by CGC/MS SIM. The results suggested that the nuclear DNA damage in cells caused by H 2O 2 may be involved in formation of hydroxyl radicals.
