胎盘MT1及ZnT1在低锌致胎鼠心脏畸形的表达

Significances of expressions of MT1 and ZnT1 in placenta of rat embryo with cardiac anomalies induced by zinc deficiency

摘要 目的 探讨胎盘金属硫蛋白-1及锌转运蛋白-1在低锌诱导大鼠胚胎心脏发育畸形中的作用及可能机制.方法 SD成年健康清洁级雌鼠50只,随机分为:极低锌、低锌、中等低锌、边缘低锌及常锌组.分别饲以不同含锌量饲料.常规方法 交配受孕,于孕19天开胸抽取孕鼠下腔静脉血检测血锌值;剖取胚心行胎鼠心脏组织学观察;RT-PCR法半定量测定胎盘中金属硫蛋白-1及锌转运蛋白-1.结果 各低锌组孕鼠血锌值明显低于常锌组(均P<0.05);极低锌、低锌及中等低锌组较常锌组胚心畸形率明显增加(均P<0.05);极低锌、低锌及中等低锌组胚鼠胎盘金属硫蛋白-1及锌转运蛋白-1 mRNA表达较常锌组明显降低(均P<0.05);心脏畸形胚鼠胎盘金属硫蛋白-1及锌转运蛋白-1 mRNA表达较正常胚鼠明显降低(均P<0.05).结论 中等以上低锌孕鼠胚心畸形率明显增加,可能与胎盘金属硫蛋白-1及锌转运蛋白-1 mRNA表达下降有关. Objective To explore signficances of expressions of metallothionein-1 (MT1) and zinc transporter-1 (ZnT1) mRNA in placenta of rat fetus with cardiac anomalies induced by zinc deficiency and their possible mechanisms. Methods 50 SD-heahhy adult female rats (body weight: 200-250g) were randomly divided into 5 groups: zinc extremely deficient group, zinc deficient group, zinc moderately deficient group, zinc marginally deficient group, normal zinc group. The rats were fed with forages with different contents of zinc. They were mated with male rats. All the pregnant rats were killed on the 19th day of pregnancy, the embryonic heart was dissected and cardiac anomalies were observed histologically. The blood samples were taken from inferior vena cava of pregnant rats for detection of zinc by using atomic absorption spectrophotometer. The expressions of MT1 and ZnT1 mRNA in placenta were detected by using semi-quantitative RT- PCR method. Results As compared with normal zinc group, the blood zinc contents in all the zinc deficient groups were significantly lower ( all P 〈 0. 05 ). The incidences of embryonic heart malformations in zinc extremely deficient group, zinc deficient group and zinc moderately deficient group were all significantly higher than that in the normal zinc group ( all P 〈 0.05 ). Also, the expressions of MT1 and ZnT1 mRNA in placenta of pregnant rats with embryonic heart malformations in zinc extremely deficient group, zinc deficient group and zinc moderately deficient group were all lower than those in the normal zinc group ( all P 〈 0.05 ). Conclusion The incidence of embryonic heart malformations in the moderately and over zinc deficient rats is significantly increased. This may be related to down-regulated expressions of MT1 and ZnT1 mRNA in placenta.
