摘要
目的运用傅立叶变换红外光谱仪和显微共焦拉曼光谱仪进行光谱测量,探讨宫颈癌组织与宫颈重度上皮内瘤变(CINⅢ)组织的光谱特征,为光谱测量在早期诊断宫颈癌中的使用提供临床依据。方法 2009年8~12月于我院就诊的宫颈鳞癌及CINⅢ患者各20例,患者经手术切除或活检获得病变组织,均利用傅立叶变换红外光谱仪和显微共焦拉曼光谱仪,分别进行红外光谱和拉曼光谱的测定。取其相应病变旁正常组织作为对照,也进行上述光谱的测定。被检测的组织相应分为四组:宫颈鳞癌组织20例,宫颈鳞癌旁组织20例,CINⅢ组织20例,CINⅢ旁组织20例。结果宫颈癌病变组织红外光谱和拉曼光谱与癌旁正常组织、CINⅢ组织、CINⅢ旁组织的光谱存在明显差异。红外光谱显示:除宫颈癌组织外,其余三种组织的光谱外观相似。宫颈癌组织具有特征性的971cm-1峰位,而CINⅢ组织、CINⅢ旁组织及癌旁组织,在971cm-1附近未检测到峰的出现。拉曼光谱显示:CINⅢ旁组织和癌旁组织光谱极为相似,宫颈癌组织与之不同,1639cm-1处振动峰为宫颈癌组织特征峰,CINⅢ组织和癌旁组织在1639cm-1附近未检测到峰的出现。结论癌组织的红外光谱和拉曼光谱具有特征峰。傅立叶变换红外光谱和拉曼光谱仪有望用来区分宫颈癌病变与正常宫颈组织,成为早期诊断宫颈癌的新方法。
Objective To use FTIR spectrum and Raman Spectrum technology was performed on cervical invasive squamous cell carcinoma tissues and cervical intraepithelial neoplasia Ⅲ(CIN Ⅲ), as well as their surrounding normal tissues.Methods 20 samples of cervical invasive squamous cell carcinoma tissues and cervical intraepithelial neoplasia Ⅲ (CIN Ⅲ) tissues and normal tissue surrounding them were taken during surgery or direct biopsy from the Second Affiliated Hospital of Sun Yat-Sen University from August 2009 to December 2009.FTIR spectrum and Raman spectrum technology were used to investigate the cervical invasive squamous cell carcinoma tissues and cervical intraepithelial neoplasia Ⅲ (CINⅢ) tissues, as well as their surrounding normal tissues.Samples were divided into 4 groups, each group including 20 samples.Results FTIR results showed that apart from the spectral band of cervical cancer tissues, other three bands are almost similar.The cervical cancer tissues showed a specific band of 971 cm-1 range, and the other groups did not show that band.In Raman Spectrum analysis the result showed that tissues around CINⅢ and tumor are relatively normal; cervical cancer tissues were different, a 1 639 cm-1 band was only observed in cervical cancer tissues.The band could not be detected in other tissues.Conclusion The cervical cancer tissues show a specific band in FTIR and Raman.The Raman spectrum result of lesion tissue was obviously different,in both molecular composition and structure.Therefore using the spectroscope to diagnose the lesion tissue and normal cervical tissue is a new method to diagnose the cervical cancer early stage.
