摘要
目的:研究选择性COX-2抑制剂NS-398对人卵巢癌细胞株SW626细胞Snail、MMP-2表达的影响,探讨选择性COX-2抑制剂NS-398对人卵巢癌细胞株SW626细胞侵袭力的影响机制。方法:以不同浓度的NS-398作用于人卵巢癌细胞,MTT法测定NS-398药物的浓度对SW626细胞的生长抑制作用,实时荧光定量PCR法分别检测NS-398作用于人卵巢癌细胞株SW626的Snail以及MMP-2的mRNA的表达水平,免疫细胞化学在蛋白水平定性检测Snail以及MMP-2的表达情况。结果:MTT法测定结果显示:NS-398对SW626细胞有明显的生长抑制作用,并呈剂量依赖性和时间依赖性;RT-PCR半定量结果显示:NS-398可使人卵巢癌细胞株SW626Snail蛋白和MMP-2的表达明显减弱,且呈剂量梯度下降,呈浓度依赖关系(P<0.05),各实验组相比较有统计学意义(P<0.05)。结论:选择性COX-2抑制剂NS-398能够抑制人卵巢癌细胞株SW626的Snail蛋白和MMP-2表达,使Snail以及MMP-2表达下调,这可能是其抑制人卵巢癌细胞株SW626肿瘤细胞侵袭力的机制之一。
Objective:To research the effect of selective COX-2 inhibitor NS-398 on expressions of Snail and MMP-2 in human ovarian cancer cell line SW626 cells,explore the mechanism of effect of NS-398 on invasive ability of human ovarian cancer cell line SW626 cells.Methods:Human ovarian cancer cells were treated with various concentrations of NS-398,MTT method was used to detect the inhibitory effect of different concentrations of NS-398 on growth of SW626 cells;real-time fluorescence quantitative PCR method was used to detect the expression levels of Snail and MMP-2 mRNA in human ovarian cancer cell line SW626 cells after treated with NS-398;immunocytochemical method was used to detect the expression levels of Snail and MMP-2.Results:The results of MTT method indicated that NS-398 inhibited the proliferation of SW626 cells,showing dose-dependence and time-dependence;RT-PCR semi-quantitative results showed that NS-398 reduced the expressions of human ovarian cancer cell line SW626 Snail protein and MMP-2 significantly,showing dose-dependence(P0.05),there was significant difference among different groups(P0.05).Conclusion:Selective COX-2 inhibitor NS-398 can inhibit the expressions of Snail protein and MMP-2 in human ovarian cancer cell line SW626 cells,down-regulate the expressions of Snail protein and MMP-2,which may be one of the mechanisms of inhibiting invasive ability of human ovarian cancer cell line SW626 cells.
