摘要
目的研究转录因子激活蛋白-2α(AP-2α)的表达在大肠癌发生发展过程中的作用。方法以60例大肠癌手术标本为研究对象,采用实时荧光定量聚合酶链反应(PCR)法分别检测大肠癌和癌旁正常组织中AP-2α基因的mRNA表达水平;以60例大肠癌组织AP-2αmRNA的表达均值为界限,将大肠癌组织AP-2αmRNA表达分为高、低表达,比较不同临床资料、病理参数下大肠癌组织AP-2αmRNA的表达率。结果大肠癌组织中AP-2αmRNA表达水平显著低于癌旁正常组织(P<0.01);大肠癌组织AP-2αmRNA低表达率在不同性别、年龄、肿瘤部位患者差异均无统计学意义(P>0.05),但其表达降低或缺失与大肠癌的组织学分级、Duke's分期及淋巴结转移密切相关(P<0.05,P<0.01)。结论转录因子AP-2α在大肠癌的发生发展过程中起重要作用,可能成为大肠癌防治的一个新靶点。
Objective To study the effects of transcription factor activator protein-2α(AP-2α)in the process of occurrence and development of colorectal cancer.Methods AP-2α mRNA expression was examined in 60 colorectal cancer samples and their paraneoplastic normal tissues by Real-Time PCR technique.Colorectal cancer tissues were divided into AP-2α mRNA high expression group and low expression group based on the average value of all the 60 patients.The expression rate of AP-2α mRNA in colorectal cancer tissues with different clinical features and pathological parameters,were compared.Results AP-2α mRNA expression in the colorectal cancer tissues was significantly lower than that in the paraneoplastic normal tissues(P〈0.01).AP-2α mRNA expression rates of colorectal cancer tissues showed no significant difference in different clinical features for sex,age and tumor site(P〈0.05).But the decrease and loss of expression rates of AP-2α mRNA were obviously related with the different histological grade of colorectal cancer,Dukes stages and lymph node metastasis(P〈0.05,P〈0.01) Conclusion AP-2α plays an important role in the process of occurrence and development in colorectal cancer,and it may become a new target for the prevention and cure of colorectal cancer.
