用单细胞凝胶电泳技术评价五指毛桃对辐射小鼠DNA损伤修复作用 被引量:2

摘要 目的探讨五指毛桃对辐射小鼠组织细胞DNA损伤的修复作用。方法 6 Gy60Coγ射线全身一次性照射建立辐射损伤模型,3个低、中、高给药+辐射组分别灌胃给予相当于生药5、10、20 g.kg-1.d-1五指毛桃水提液,每天1次,连续7 d,阴性对照组、单纯辐射组给予生理氯化钠溶液,用单细胞凝胶电泳技术检测小鼠组织细胞DNA损伤和修复情况。结果各辐射+给药组小鼠组织细胞DNA的尾部DNA百分率(Tail DNA%)和尾矩(Tail moment)均低于单纯辐射组(P<0.01),且随着给药剂量的增加,Tail DNA%和Tail moment越低。结论五指毛桃对辐射小鼠各组织细胞DNA的损伤有修复作用,具有一定的抗辐射作用。 Objective To explore the effect of DNA damage repair induced by Radix Fici Hirtae in irradiated mice.Methods The mice model of radiation damage was established by single whole body irradiation at the dose of 6 Gy with 60Co γ-rays.Groups at low,medium and high doses plus radiation were respectively given aqueous extracts of Radix Fici Hirtae at 5,10 and 20 g·kg^-1·d^-1 for 7 consecutive days.The control group and radiation model group were given normal saline.DNA damage and repair were detected by single cell gel electrophoresis.Results Tail DNA% and tail moment of tissue cells of the radiation plus dose groups were lower than those of the radiation model group(P〈0.01).With the doses increased,tail DNA% and tail moment were reduced.Conclusions Radix Fici Hirtae can repair DNA damage induced by radiation in mice and has some anti-radiation effects.
