摘要
以文心兰的侧芽为外植体,对侧芽的选择、处理,外植体的消毒方法、培养基的选择等技术进行研究。结果表明:最适合的材料为叶片还未展开的侧芽,二次消毒对文心兰侧芽消毒效果最好。文心兰理想的诱导培养基为MS+6-BA 5.0 mg/L+NAA 0.5 mg/L+椰子水100 mL/L,增殖培养基为MS+6-BA 2.0 mg/L+NAA 0.2mg/L+椰子水100mL/L,壮苗培养基为MS+香蕉20g/L,生根培养基为1/2MS+NAA1.0mg/L+香蕉50g+土豆20g/L+AC2.0 g/L。同时通过对炼苗时间、移栽前的处理及移栽后管理方法的综合研究,摸索出一套适合文心兰移栽的方法,移栽成活率达到90%以上。
The lateral buds of oncidium were used as explants. The effects of material, disinfecting methods and culture media on in vitro growth of oncidium were investigated. The results were as follows: the best material was the lateral buds whose blades did not unfold. Two times of consecutive disinfecting attained the best result. The most appropriate culture media were as follows: induction medium MS+6-BA 5.0 mg/L+NAA 0.5 mg/L+coconut water 100 mL/L; proliferation medium MS+6-BA 2.0 mg/L+NAA 0.2 rag/L+ coconut water 100 mL/L; seedling medium MS+banana 20 g/L; rooting medium 1/2MS+NAA 1.0 mg/L+ bananas 50 g/L+potato 20 g/L+AC 2.0 g/L. The transplanting survival rate was over 90% after pretreatment and management of oncidium plantlets.
