摘要
目的:运用RP-HPLC法建立绿原酸分析方法。方法:采用Waters 1525液相色谱附二极管阵列检测器与紫外检测器,Waters ODS C18柱(3.9 mm×150 mm,4μm),流动相:2.5 mmol/L四丁基溴化铵水溶液-乙腈(87∶13),检测波长:325 nm,流速:1 mL/min;柱温:25℃。结果:绿原酸线性范围为0.41-41μg/mL,r=0.999 9,样品平均回收率为99.81%,RSD为1.49%。结论:本方法简便、准确可靠,适用于松针叶及成分复杂且含量低的常绿植物中绿原酸含量测定。
Objective: To establish an ion pair chromatography method for the determination of chlorogenic acid of antibiotic active substance in evergreen plants.Methods: The determination was performed on a Waters C18 mm ODS column(4 μm,3.9mm×150 mm)and mobile phase consisted of 2.5 mmol/L tetrabutyl ammonium bromide-acetonitrile(87∶13,v/v),the detection wavelength was 325 nm,the flow rate was 1 mL/min and the column temperature was set at 25 ℃ by Waters 1525 HPLC with diode-array detector and ultraviolet absorption detector.Result: The method had good linear relationship within the range of 0.41-41 μg/mL(r=0.999 9).The average recovery was 99.81%(RSD=1.49%).Conclusions: The method is simple,reliable and accurate for the determination of chlorogenic acid with complicated component and low content in evergreen plants.It will provide a scientific basis for the developing and utilizing of pinus needle of Tabulaeformis Carr as medicinal natural resources in Gansu.
