摘要
Nicastrin(NCT)是高度糖基化的I型跨膜蛋白,是γ-分泌酶复合物的重要组件蛋白之一,广泛分布于人类或鼠的所有细胞类型。它不仅与γ-分泌酶的组装和成熟密切相关,其构象及表达变化对γ-分泌酶活性和阿尔茨海默病(Alzheimer's disease,AD)中β淀粉样蛋白(amyloid proteinβ,Aβ)的产生及降解也起重要调节作用。本文就国际上近几年在NCT的结构、合成、分布、降解及功能等方面的研究进展作一综述。
Nicastrin(NCT) ,one of the most important components of γ-secretase,is a highly glycosylated type I transmembrane glycoprotein. It is expressed in almost all cell types in human and mouse. NCT is not only closely related to the assembly and maturation of γ-secretase complex,more importantly,changes of its conformation and expression have a significant effect on the regulation of γ-secretase activity,as well as the generation and degradation of amyloid β peptide in Alzheimer's disease. This review summarizes the development of structure,synthesis,distribution,degradation and function of NCT in recent years,including that in our laboratory.
