摘要
以兰州百合的内层鳞片为外殖体,以MS为基本培养基并添加不同配比生长调节剂,进行组织培养研究。结果表明,以MS+1.0 mg/L 6-BA+0.2 mg/L NAA培养基对初代培养形成不定芽最佳,低温有助于外殖体分化;不定芽增殖最适培养基为MS+0.5 mg/L 6-BA+0.5 mg/L NAA;1/2 MS+0.2 mg/L NAA+蔗糖60 g/L适宜苗的生根培养,生根率达100%,且结鳞茎率达73.4%。
The inner flakes of lily were used as explants to induce the additional shoots in MS medium with different hormone combinations,and the effects of hormones and temperature on the induction of shoots were studied.The results showed that the best culture medium for the induction and differentiation of addition shoot was MS + 1.0 mg/L 6-BA + 0.2 mg/L NAA in low temperature;the best medium in the stage of shoots multiplications was MS + 0.5 mg/L 6-BA + 0.5 mg/L NAA;and 1/2 MS + 0.2 mg/L NAA + sucrose 60 g/L was the most effective media for rooting inducement,its rooting rate was 100% and the bulb rate was 73.4%.
