摘要
目的通过双向电泳技术对比研究变形链球菌标准株与LuxS突变株菌体总蛋白质的表达差异,为进一步研究LuxS信号系统对变形链球菌致龋毒力的调控机制奠定基础。方法提取变形链球菌标准株及LuxS突变株的菌体总蛋白质,采用双向电泳技术对比研究两者蛋白质的表达差异,找出差异蛋白点。结果与标准株菌体总蛋白2-DE图谱相比较,LuxS突变株菌体总蛋白2-DE图谱中有61个斑点的蛋白质表达量发生了改变:36个点蛋白质表达量升高,25个点蛋白质表达量降低。结论变形链球菌标准株与LuxS突变株菌体总蛋白存在明显的差异,表现为2-DE图谱蛋白斑点染色的深浅不同,提示LuxS突变株菌体总蛋白发生了量的变化。
Objective Byusingtwo-dimensional electrophoretic techniques,to compare the protein expression differences of two strains,which can give a significant support to the further study of the signal system of LuxS mutant,whose adjustment mechanismcan cause dental caries. Methods Fortwo-dimensional electrophoretic analysis,in order to compare the expression differences and find out the difference points by using two-dimensional electrophoretic technique the total protein of the standard strain and LuxS mutant were extracted. Results Compared with the 2-DE atlas of the standard strain,the expression quantity of 61 protein points changed in atlas of LuxS mutant,36 of which were stepped up and 25 were cut down. Conclusion There were significant differences in the total protein between the standard strain and LuxS mutant. The difference of the protein point staining shade in 2-DE atlas suggested that there was a quantitative alleosis of the total protein in LuxS mutant.
