伏翼DNA指纹谱探针的筛选

摘要 以人工合成的微卫星序列(GTG)5、(GT)8、(CAC)5和人源小卫星33.15做引物,首次随机扩增了伏翼的基因组DNA,从多态性DNA片段中回收8条特异性片段,即(GTG)5a、(GTG)5b、(GT)8a、(GT)8b、(CAC)5a、(CAC)5b、33.15a和33.15b.与被标记过的基因组DNA做探针斑点杂交结果表明,8个片段中(GTG)5a、(CAC)5a和33.15a产生了阳性信号,因此,这三个片段被标记后做探针可用于伏翼DNA指纹谱的制备. The DNA polymorphic fragments of Pipistrellus abramus were first amplified by the polymerease chain reaction (PCR) with artificial primers that contained (GTG)5,(GT)8,(CAC)5 and human minisatellite 33.15. Eight bands were retrieved among the DNA electrophoresis polymorphic bands.it were (GTG)a、(GTG)b、(GT)a、(GT)b、(CAC)a、(CAC)b、33.15a and 33.15b.The results of dot hybridization showed that, When the eight fragments and Genomic DNA as the probes were hybridized,(GTG)a、(CAC)a and 33.15a could emerge positive signals.It means that the three fragments labeled could be applied in the analysis of Pipistrellus abramus DNA fingerprint.
