RNA干扰沉默STAT3基因表达对人大肠癌细胞生物学行为的影响 被引量:3

摘要 目的探讨RNA干扰(RNAi)技术沉默STAT3基因表达对人大肠癌细胞生物学行为的影响。方法采用真核细胞转染技术将pSi-lencer3.0-H1-siRNA-STAT3重组质粒转染人结肠癌HCT116细胞,MTT法检测细胞增殖,RT-PCR、Western blot及免疫组化法分别观察STAT3基因和蛋白的变化,流式细胞仪及AO/EB染色观察细胞凋亡。结果 pSilencer3.0-H1-siRNA-STAT3重组质粒对大肠癌细胞的增殖有明显抑制作用,与空白组及阴性组相比差异有统计学意义(P<0.05);在重组质粒组,STAT3基因mRNA及蛋白的表达与空白组及阴性组相比明显减低(P<0.05);重组质粒可诱导HCT116细胞凋亡,细胞周期分期示细胞阻滞于G2期。结论 pSilencer3.0-H1-siRNA-STAT3重组质粒可抑制STAT3基因在人大肠癌细胞中的表达。
