摘要
采用改良的PCRRFLP方法测定长沙地区291名儿童载脂蛋白(Apo)E基因型。结果发现ε2,ε3,ε4等位基因的频率分别是0081,0857和0062。ApoE等位基因频率分布无明显的性别差异(P>0.05)。Apoε2等位基因携带者与其它ApoE基因携带者相比,血高密度脂蛋白胆固醇、ApoE和ApoA1浓度较高,而总胆固醇、低密度脂蛋白胆固醇、ApoB100水平较低(P<0.05)。ε4等位基因携带者的血脂、脂蛋白、Apo改变则相反(P<0.05)。结果还显示ApoE等位基因对血甘油三酯、脂蛋白(a)浓度的影响不明显(P>0.05)。ε2,ε3,ε4等位基因与血总胆固醇、低密度脂蛋白胆固醇、ApoB100浓度呈等级正相关,与高密度脂蛋白胆固醇、ApoA1及ApoE水平呈等级负相关(P<0.05)。ApoE基因对血清脂类的影响存在性别差异。ApoE基因多态性影响小儿血清脂类代谢。
In order to investigate the distribution of apolipoprotein(Apo) E polymorphism and effect of ApoE genotypes on plasma lipid, lipoprotein, apolipoprotein levels in children of Changsha, we determined ApoE genotypes in 291 children by means of PCR RFLP. ApoE allele frequencies in the population were ε 2=0.081, ε 3=0.857, ε 4=0.062. There were no difference in the ApoE genotypes distribution between sexes. The children with Apoε 2 allele had higher levels of ApoE, ApoA 1 and high density lipoprotein cholesterol together with lower levels of total cholesterol, ApoB 100 and low density lipoprotein cholesterol than those with other ApoE alleles. These changes were opposite in children with Apoε 4 allele. The levels of plasma triglyceride, lipoprotein(a) were not affected significantly by ApoE alleles. Moreover, the effect of ApoE genotypes on levels of plasma lipids was different between sexes. These results suggest that ApoE polymorphisms could affect plasma lipids metabolism in children.
