摘要
目的研究白芍总苷(TGP,抗变态反应药)对大鼠胶原性关节炎(C IA)及足爪组织MMP-9表达的影响。方法用鸡Ⅱ型胶原建立C IA大鼠模型,观察药物对C IA大鼠体质量及足肿胀度的影响,用免疫组化法观察大鼠足爪组织中MMP-9蛋白的表达。结果 TGP可有效缓解C IA大鼠体质量减轻,明显抑制C IA大鼠足爪肿胀;大、中剂量TGP组的足爪组织MMP-9阳性表达较模型对照组明显降低,有非常显著性差异(P<0.05或P<0.01)。结论 TGP可明显改善C IA大鼠症状,其机制可能与其下调MMP-9表达、降低血清IL-1β水平有关。
Objective To explore effects of total glucosides of paeonia (TGP) on the body weight, degree of foot swelling and the matrix metal- loproteinase- 9 (MMP- 9 ) protein expression in paws tissues in colla- gen - induced arthritis (CIA) rats. Methods CIA model in SD rats was established. The effect of drugs on the body weight and the degree of foot swelling of the CIA rats was to observed. The expression of MMP - 9 proteins in rat, spaw tissues was detected by immunohistochemical staining. Results TGP could effectively relieve the lighten of the body weigh and inhibit the paw's swelling degree of the CIA rats. The MMP -9 expression was significantly lower in large and medium -dose TGP than in the model group, the difference was significant (P 〈 0. 05, P 〈 0. 01 ). The positive expression of MMP -9 in low -dose group, TGP had no significant differ- ence compared with the model group. Conclusion The therapy effect of TGP maybe related to decreasing the expression of MMP -9 protein and suppressing the production of IL - 1 β in CIA rats.
