摘要
目的研究紫杉醇(抗癌药)对成纤维细胞基质金属蛋白酶-2(MMP-2)和α-平滑肌肌动蛋白(α-SM-actin)的影响。方法选取雄性SD大鼠,用贴壁法培养胸主动脉外膜成纤维细胞(AF),用RT-PCR方法检测α-SM-actin和MMP-2。结果成纤维细胞的MMP-2的表达量,紫杉醇组较对照组也明显降低(P<0.01);α-SM-actin的表达量,紫杉醇组较对照组明显降低(P<0.05),表明紫杉醇能抑制AF的MMP-2和α-SM-actin的表达,抑制AF的表型转化,从而抑制血管外膜的迁移。结论紫杉醇可防治冠状动脉介入术后再狭窄。
Objective To investigate the effect of paclitaxel on the ex- pression of matrix metalloproteinases - 2 ( MMP - 2 ) and α - smooth muscle aetin ( α- SM - actin) of the fibroblast. Methods Adventitial fibroblast (AF) were prepared from rat aortas using explant technique and cultured for passages, the expressions of MMP - 2 and α - SM - actin were determined by RT - PCR. Results The expression of MMP - 2 in the paclitaxel group is significantly lower than that in the control group (P 〈 O. 01 ). The expression of α - SM - aetin in the paclitaxel group is signif- icantly lower than that in the control group( P 〈 0. 01 ). Paclitaxel can in- hibit the expression of MMP - 2 and α - SM - actin of the fibroblast, ef- fectively inhibiting the phenotype transformation of AF, thus inhibiting the multiplication and migration of vascular adventitia. Conclusion Paclita- xel can prevent restenosis after percutaneous coronary intervention.
