摘要 探讨 P C R- S S C P- 银染法在基因突变检测中的应用. 应用 P C R- S S C P- 银染法分析了苯丙酮尿症患者的 P A H 基因外显子4 , 7 , 10 , 11 和12 , 并对部分样品进行 D N A 测序. 结果: 检出部分已知突变如 R243 Q, R413 P 等, 并发现当 S S C P 带型不同于阳性、阴性对照时, 或是外显子其它突变, 或是内含子碱基变化引起, 也可见于碱基序列正常的样品中. 结论: P C R- S S C P- 银染法在有较好对照, 严格控制影响因素条件下, 简便、实用, 是多样品筛查、 D N A 测序前筛查的较好方法. To explore application of PCR-SSCP-silver stain method in gene mutation analysis, exon 4,7,10,11 and 12 of phenylalanine hydroxylase gene in phenylketonuria were analysed by PCR-SSCP-silver stain method. Results: Partly known mutations, i e. R243Q and R413 were identified. Other mutations in exons, base alteation in intron or some samples in normal DNA sequence cause bands of SSCP differing from positive and negative contrast. Conclusion: With better contrast and strictly controlling influence factors, PCR-SSCP-silver stain method is a simple, practical and better method for screening multiple samples and DNA sequencing.
