抗生素对沂州木瓜离体叶片不定芽再生的影响 被引量:3

摘要 [目的]研究卡那霉素(Kan)、头孢霉素(Cef)和羧苄青霉素(Carb)对木瓜幼芽嫩叶不定芽再生的影响。[方法]采取沂州木瓜培养新梢嫩叶处理,依次接种在再生培养基及生根培养基上,注入设计浓度的对应抗生素,分析3种常见抗生素Cef、Kan、Carb对沂州木瓜培养新梢嫩叶上不定芽产生的效果。[结果]沂州木瓜对Kan反应极强烈,浓度为5mg/L时,叶片不定芽再生率非常低(2.4%);浓度为10mg/L时,芽分化被完全抑制,叶柄略有膨大,组织变白,有微量愈伤组织;浓度为15~30mg/L时,叶片既不能形成愈伤组织也不能分化不定芽。Cef和Carb对沂州木瓜叶片生根及生长的作用呈负相关关联,浓度在100mg/L时均同对照差异很小,随着添加浓度的升高,抑制作用则显著表现出来,500mg/LCef或400mg/LCarb即可阻止沂州木瓜新梢嫩叶的愈伤组织出现。[结论]沂州木瓜叶片对Kan敏感,Cef和Carb对木瓜离体培养的影响相似,但Carb抑制再生的作用比Cef强烈。 [Objective] The effect of kanamycin(Kan),cefotaxime(Cef)and carbenicillin(Carb)on the adventitious bud regeneration from papaya young leaf-culturing in vitro.[Method] The fresh young papaya leaf,after it was pretreated,was firstly cultured in the regeneration medium and then the rooting medium,which were inoculated with the corresponding concentration of antibiotics;and the efficiency of three kinds of common antibiotics:Cef,Kan and Carb,affecting adventitious bud regeneration was analyzed.[Results] The sensitivity of the Yizhou papaya leaf culturing to Kan was very strong,the regeneration rate of adventitious bud was very low(2.4%)while its concentration was 5 mg/L;the bud differentiation was completely inhibited,only with slightly swollen petiole with white tissue and a little callus-forming while its concentration was 10 mg/L;and both the callus from leaf and the differentiation of leaf never appeared while its concentration was 15-30 mg/L.There was negative correlation between the Cef/Carb and the papaya leaf growth and its tissue rooting.At the concentration of 100 mg/L,the treating efficiency of Cef and Carb was almost same as that of the CK;and the inhibition of the treatments to its growth was significantly improved with the increment of Cef/Carb's concentration,at the concentration of Cef of 500 mg/L or Carb of 400 mg/L,the formation of callus from tender leaf was inhibited.[Conclusion] Yizhou papaya leaf was sensitive to Kan.The impact of Cef and Carb on papaya culture in vitro was similar but the inhibition of Carb to the regeneration of the tissue from Yizhou papaya leaf culturing was stronger than that of Cef.
