摘要
[目的]为黄色鸡冠花规模化生产提供依据和参考。[方法]用黄色鸡冠花的种子进行无菌萌发,以MS为基本培养基,附加不同浓度的激素组合进行组织培养与快繁技术研究。[结果]黄色鸡冠花种子的灭菌采用浓度75%乙醇表面消毒60s和0.1%HgCl2消毒8min的处理方式效果最好;在MS+6-BA1.0mg/L+NAA0.050mg/L培养基上可以很好地诱导产生愈伤组织,产生丛生芽的数量最多;生根的最适培养基为:1/4MS+IBA0.3mg/L,生根率达93%。[结论]该组织培养体系适合应用于生产。
[Objective] The aim was to provide a basis and reference for large-scale production of yellow Celosia cristata L..[Method] Research on tissue culture and rapid propagation in MS basic medium supplemented with different concentrations of hormone after seeds sterile germination.[Result] The best treatment of seed surface sterilization was with 75% ethanol for 60 seconds and 0.1% HgCl2 for 8 minutes.Callus could be induced well and resulted in the largest number of rosette buds in the MS medium with 6-BA 1.0 mg /L and NAA 0.050 mg/L;the best medium for rooting is that 1/4MS with IBA0.3 mg/L,and the rooting rate was 93%.[Conclusion] This tissue culture system is suitable for production.
