摘要
目的:建立HPLC法测定肺舒通喷雾剂(金荞麦、丹参、苦杏仁、麻黄、甘草)中(-)-表儿茶素、丹参酮ⅡA和丹酚酸B含量的方法。方法:采用HPLC法对制剂中的3种组分分别测定,以Symmetry shield RP C18色谱柱(4.6mm×250mm,5μm)为固定相,(-)-表儿茶素:流动相:0.05%三氟乙酸水溶液-乙腈(9∶1),检测波长:280nm,流速:1.0mL/min,柱温:40℃。丹参酮ⅡA:流动相:甲醇-水(75∶25),检测波长:270nm,流速:1.0mL/min,柱温:35℃。丹酚酸B:流动相:甲醇-乙腈-水-甲酸(30∶10∶59∶1),检测波长:286nm,流速:0.8mL/min,柱温:35℃。结果:(-)-表儿茶素在1.7438~27.9000μg/mL、丹参酮ⅡA在0.5125~8.2000μg/mL,丹酚酸B在6.275~100.400μg/mL范围内具有良好的线性关系,相关系数r分别为0.9999、0.9998和0.9998;平均加样回收率分别为101.03%(RSD=2.24%)、100.55%(RSD=1.30%)和99.99%(RSD=2.10%)。结论:本方法专属性强,结果准确,重复性好。
AIM: To establish the methods for determining (-)-epicatechin,tanshinoneⅡA and salvianolic acid B in Feishutong Spray( Rhizoma Fagopyri dibotryis,Radix et Rhizoma Salviae miltiorrhizae,Semen Armeniacae ama-rum,Herba Ephedrae,Radix et Rhizoma Glycyrrhizae) by HPLC. METHODS: Symmetry shield RP C18 column (4. 6 mm × 250 mm,5 μm) was used. The mobile phase for (-)-epicatechin was 0. 05% trifluoroacetic acid-acetonitrile(9 ∶ 1) at the flow rate of 1. 0 mL/min. The column temperature was 40 ℃ and detection wavelength was set up at 280 nm. The mobile phase for tanshinoneⅡA was methanol-water(75 ∶ 25) at the flow rate of 1. 0 mL/min. The column temperature was 35 ℃ and detection wavelength was set up at 270 nm. The mobile phase for salvianolic acid B consisted of methanol-acetonitrile-water-formic acid (30 ∶ 10 ∶ 59 ∶ 1) at the flow rate of 0. 8 mL/min. The column temperature was 35 ℃ and detection wavelength was set up at 286 nm. RESULTS: The linear ranges of (-)-epicatechin,tanshinoneⅡA and salvianolic acid B were in the ranges of 1. 743 8-27. 900 0 μg/ mL(r = 0. 999 9),0. 512 5-8. 200 0 μg/mL (r = 0. 999 8) and 6. 275-100. 400 μg/mL (r = 0. 999 8),respec-tively. The average recoveries were 101. 03% (RSD = 2. 24% ),100. 55% (RSD = 1. 30% ) and 99. 99% (RSD = 2. 10% ),respectively. CONCLUSION: The methods are accurate,sensitive,reproducible and suitable for the quality control of Feishutong Spray.
