摘要
目的:探讨七福饮对海马注射β-淀粉样蛋白(Aβ1-42)的大鼠海马神经细胞凋亡的保护作用及机制。方法:84只SD健康大鼠随机分为空白对照组、假手术组、模型组、西药组、七福饮低剂量、七福饮中剂量和七福饮高剂量组,凝聚态Aβ1-42大鼠海马定向注射诱导老年性痴呆(Alzheime's disease,AD)动物模型,采用免疫印迹法检测大鼠脑组织Bcl-2和Bax蛋白的表达,透射电镜观察海马组织超微结构的变化。结果:与假手术组比较,模型组Bax蛋白表达相对值显著升高,而Bcl-2相对值显著降低(P<0.05);与模型组比较,西药组、七福饮低、中、高各剂量组Bax蛋白表达相对值显著降低(P<0.05),而七福饮低、中、高各剂量组Bcl-2蛋白表达相对值显著升高(P<0.05)。电子显微镜观察结果表明,模型组大鼠海马变性,七福饮能改善大鼠海马神经元病理改变。结论:七福饮可通过调节海马组织Bax和Bcl-2基因的表达水平、抑制神经细胞凋亡的发生,而对AD大鼠损伤的神经细胞起到保护作用。
Objective:To observe the protective effect of Qifu Yin on the apoptosis in hippocampal neurons in Alzheimer's disease(AD) rats induced by amyloid-β1-42 injection.Method: Eighty-four SD rats were equally randomly divided into blank control group,sham controls group,model control group,donepezil control group and Qifu yin groups(2.15,4.3,8.6 mg.kg-1).The AD model was established by amyloid-β1-42 injection into the bilateral hippocampus.Expression of Bax and Bcl-2 protein in rat brain was estimated by western blotting method.Change of hippocampus is observed by electron microscope.Result:The expression of protein Bax was up-regulated in model group compared with the sham group and Bcl-2 was down-regulated.In Qifu Yin group,the Bax expression was down-regulated and Bcl-2 up-regulated compared with the model group(P 0.05).Qifu Yin can alleviate the damage of hippocampus neurons.Conclusion:Amyloid-β1-42 injection into the bilateral hippocampus can up-regulate expression of protein Bax and down-regulate expression of Bcl-2 in brain tissue.Qifu Yin inhibits apoptosis through regulating the expression of Bcl-2 and Bax protein in hippocampal.may provide some protective effects on cerebral neurons damage of AD model in rats.
